Germplasm Phenotype:

  • bin4-1  — Various organs of this brassinolide-insensitive dwarf mutant, such as leaves and roots, are only about 25% the size of wild-type organs. Their cells have smaller nuclei and reduced ploidy, based on analyses of 12-day old leaves. bin4-1 phenotypes are not altered in triptychon, rastafari, kaktus2, or glabra2 mutants. However, chromosome doubling induced by colchicine does lead to increased cell size. SPO11-3/RHL2/BIN5:GFP can still enter the nucleus in the bin4-1 mutant background. The transcript level of several DSB-inducible genes, such as PARP1 is higher in bin4-1 mutants, but these mutants do not show general defects in the maintenance of transcriptional gene silencing. Elevated levels of DSB-responsive transcripts in bin4-1 can be partially suppressed by atm or atr mutations. There is ectopic expression of the transcriptional CYCB1;1:GUS reporter in non-dividing cells in the bin4-1 mutant. However, the transcript levels of many other G2/M genes are not altered based on RT-PCR analysis.
  • bin4-2  — The bin4-2 mutant has a dwarf phenotype with reduced cell size and a reduction in the maximum amount of endoreduplication observed in leaf cells. Cotyledon cells from bin4-2 mutants have higher levels of double strand breaks than comparable wild-type cotyledon cells.
  • mid-1  — Small plants, reduced cell size, decreased endoreduplication, reduced number of root hairs and abnormal development of columella cells of the seed coat.
  • mid-2  — Small plants, reduced cell size, decreased endoreduplication, reduced number of root hairs and abnormal development of columella cells of the seed coat.
